I am reaching out to see what techniques do you use/recommend to validate your gene expression trends obtained through spatial transcriptomics (ST) technology (10x Visium)? (particularly in a disease context where a gene expression is up or downregulated with time).

Some say qPCR is an ideal technique for validating the could be employed to validate ST results. However, if the fold change of the gene transcript is very small (~1.2-1.5) as observed from ST data, can it be confirmed using qPCR too?

If not, why and what are the challenges? Are could you recommend any alternate technique to validate besides scRNA sequencing which seems to be expensive as compared to qPCR testing?
