I am working on my final project and using IHC as my method. But, I haven't finished my optimization especially in antigen Retrieval.

My first optimization using pressure cooker 30 minutes (the tissue is broken), the second one pressure cooker 15 minutes (the tissue is gone), the third one water bath 10 minutes (1 of the 3 slides, the tissue is broken, and the other 2 is gone).

Can you help me, what is the main problem of my method? Is it the method I use that wrong or the slides? Thank you.
