I ask basic questions like... Gender, Family Size, Family Income (Optional), Access of Internet and Smartphones, E-commerce Apps used on smartphone and only one 5 point Likert Scaled Question that "according to you, the usage of e-commerce apps has increased" 1-not at all, 5-has increased.
can I do hypothesis testing on the base of this one 5 point likert scaled question?
or shall associate the response of this scale with other variables like Family Income or Family Size, etc

I'm not a great expert but I've been in a similar situation. Yes, you can make hypotheses or establish relationships through statistical tests with the data. It may be useful for a first approach to a topic or for an academic assignment. However, if your intention is to publish in a journal, it would be difficult because the instrument isn't validated.
Having a single primary question about "[use] of e-commerce platform among ... college students ... any impact of Lockdown or not" is a weak basis for any generalisation or hypothesis testing. Remember, the other questions you mention concern so-called secondary variables, mostly used for controlling side effects produced by intervening variables, not the phenomena you are primarily interested in.
With 5-points scale responses combining individual profiles, you can do n number of tests which you need to do in line with your objectives. Best wishes.
