1. At least some parts of the leaves have GFP glowing. And the color is right (compared to those posted in another thread).
2. Why does GFP only glow on some areas (indicated by yellow arrow), not on other parts of the leaf (indicated with red circle)?
3. It is only 'empty vector' transformation. It is usually serve as a 'control'. You need a 'Gene-of-interest' in the binary vector as an 'experimental' group for publication. You might be able to get by (get published) if this is the first successful genetic transformation case in your plant species.
Some plants has autofluroscence properties that gives GFP like fluroscence. And dead cells also gives GFP like fluroscence. If you want to confirm this fluroscence are GFP signals are not, you should check either it colocalised with chlorophyll or not.
