We have prepared two cross-linked peptides having 14 Lys and 14 Asp residues. The sample is soluble in 50 % ACN-water. We are unable to get mass spectra both in ESI-MS and MALDI. We tried MALDI using DHB, alpha-cyano-4- hydroxycinnamic acid and sinnapinic acid and we used sample : matrix 1:1, 1:3 and 1:5 ratio both in positive and negative ion mode. We request for your suggestions?

What do you mean with "cross-linked"? Did you start with a 14-mer of Lys and a second peptide comprised of a 14-mer of Asp?

Hi Rituparna,
Could you please elaborate on the mode of cross-linking in your peptides ? It seems to me that you have "Isopeptide" linkage (s) in your analytes. Kindly clarify so that we can understand the problem well before resolving it.
