Can anyone recommend me the best way to culture Bifidobacterium bifidum and B. longum? I can culture B. longum on MRS agar with 0.05% cysteine under anaerobic condition using anaerocult but failed after picking colony to MRS broth with 0.05% cysteine (5 mL in 20-mL glass tube, capped) under normal atmosphere. I also tried to isolate B. bifidum from a mixed culture but never succeeded. is there any selection medium for B. bifidum from L. acidophilus and L. rhamnosus?

thank you so much. I found bifidobacteria are very sensitive to oxygen. It is best to culture them under anaerobic condition. Otherwise, limited the exposure to oxygen by smaller head space with tightly capped might help. After supplemented more cysteine to 0.0625% in MRS broth with less than 5-10% head space, B. longum started to grow. However, need to confirm no contamination occurred. :)
I guess You can help out about my Problem. I purchased ATCC Bifidum Species and started to culture. Last few days i was a little worried and tried to check if i get the right Strain or not. Normally I use MRS agar as well as MRS Broth Suplemented with 0.5 % Cystein. But Now i didnt get result on PCR. I don't know what is wrong. I use the Bug Box for culturing while using screw caped plastic tubes and then put in normal aerobic incubator for incubation. Is there any mistake in my culturing.,?
