i am working on microscopy of macrofungi but i am having a problem in the preparation of slides, can anybody please let me know which chemicals and stains to use to have a clear slide.
Have you tried with 85% lactic acid? With us it works well, e.g. for spores and observations of ectomycorrhiza. The slide can be kept for several months if sealed with nail polish (which is not very easy to do).
I'm replying a bit late, but let me tell you that your question is a bit ambiguous. Depending on the genera of macrofungi you're working on, you may need many different stains.
If your unique goal is to obtain clear slides, you can try as suggested with Lactophenol or other media as "L4 medium after Clemençon", "water with Sodium Dodecyl Sulphate", "GDS after Clemençon"...
About staining, there are also a lot of stains used in mycology. As general stain you can use Congo Red (stain walls), Phloxine (stains content), a combination of Congo Red and Phloxine (red walls and fuchsia content), Cotton blue...
But you'll need severa different stains depending on the genera of fungi you're woking with... as example only for spores... in ascomycetes Cotton blue for spore wall, for spores of Cortinarius is useful KOH, for spores of Macrolepiota Cresyl Blue, many times needed Lugol to check amiloidity...

Finally, as you surely know, most important factor in order to obtain a clear slide, is to make a good preparation, with a very thin layer of tissue.
