We are using STZ-induced diabetic mice as our model. And we are using a single dose injection, which is 180 mg/kg. I've gone through couple of literature regarding the volume of citric acid buffer, some use 0.2 ml, others use 0.4 ml, makes me wondering if it's weight-dependent. Or is there a standard concentration for the STZ solution? Any help is appreciated
When you are using Citrate buffer, it is important to keep the volume to a minimum so the pH of the blood does not change. [To avoid the pH change, you can use 1x10-4N HCl, which will have the desired pH but obviously would have no capacity to buffer.]
A decent concentration of STZ in your case will be 45mg/ml or 90mg/ml. This will make it easy to calculate the correct volume to whatever weight are your mice. For example, in the case of a 25gm mouse, you will need 100µl and 50µl, respectively.
