Regarding the highlighted part of the photo below, is the blank really the one with no catechin? How can it be so since catechin standard is added to all the test tubes (except for the 0mL aliquots)?
I thought maybe that the blank should be the one with no vanillin (this is for the construction of calibration curve not the analysis of the sample extracts themselves).
Can someone please clear this up? Thanks a lot.

The procedure what they have given is correct. Blank has to be carried out in the absence of catechin. The purpose of carrying out blank is to subtract the amount of light radiation absorbed by the reagents other than the catechin. So you have to perform blank without catechin and in the presence of vanillin.
hi Rajkumar Thangamani thank you for answering. Why, then, does the procedure say to add the catechin standards to both sets of the test tubes? If the blank needs to have no catechin, one set of test tubes need to have 0 added catechin.
Also, both sets should be added with vanillin reagent right? The procedure says that only set A tubes are added with the vanillin reagent and set B are added only with the HCl solution.
