I am searching for a solution containing both essential and non-essential amino acids. Mammalian cell culture seems to use these quite often, I would like to know if someone has experience using RPMI (all aminoacids except alanine for some unknown reason) with Bacteria cells. I have a metabolic change in my fermentations and I would like to know if aminoacids are a reason.
I have used RPMI 1640 medium to culture fungal cells for antibiotic susceptibility testing and for biofilm formation. I use the medium also for cultivation of cancer cell lines and normal mammalian cells. Therefore, I believe that eukaryotic cells could be cultured in it but prokaryotic cells may not.The medium contains no proteins, lipids or growth factors, therefore it requires supplementation.
RPMI will probably support the growth of a lot of bacteria. There are too many bacteria capable of growth in defined media containing just Glucose and mineral salts and no aminoacids or proteins, RPMI is way more richer (Contains also aminoacids and vitamins).
The questions is why to use RPMI media (Wich is expensive) instead of a defined mineral media adapted to your bacteria and supplemented with whatever aminoacid you think it's missing.....
it will probably work. However, it is quite expensive as other people have pointed out. I suggest you try CSM mixtures used for saccharomyces cerevisiae, we have used them with success for e.coli atleast and should be easier than making minimal media in which you have to add everything individually (also you have the convenience of using mixtures that have single, double, triple dropouts etc..) .
