Using raffinose to derepress the Gal promoter, does the media need glycerol as a carbon source?

I am overexpressing and purifying and enzyme from yeast. I know that I can use media with glucose for the starter culture and switch to raffinose to derepress the Gal promote and then add the galactose. With the exception of the starter culture with glucose, does the rest need glycerol as a carbon source?
