I'm trying to measure intra-tumoral metabolites using MALDI-TOF imaging (rapifleX-Bruker).
Does anyone know the methods for tumor tissue preparation?
(BEFORE mounting the tissue onto ITC glass slides!)
I'm not quite sure if fixing the tissue (w/ formaldehyde) is appropriate for such experiments....
Do I just i) extract the tumor ii) embed the tumor in proper embedding solution iii) cryosection ?
Any literature suggestions or sharing protocols would help me a lot !!!

Hi Sewon,
I would prefer to avoid any fixation and embedding if you can but it is based on the type of tissue. For tissues like brain, liver or kidney, just snap freeze the tissues in LN2, section them on cryostat and dry/fix them in dessicator. They will be fine. For some difficult tissues like lungs you might consider embedding media but avoid any paraffin or polyalcohol based media like O.C.T. or TissueTek due to signal suppression. I've seen some papers using gelatin and CMC but never tried them myself. In worse case scenario you can use paraffin and deparafinize the sections on the slide by xylene/ethanol washes but there is a risk of signal loss due to extraction into xylene/ethanol.
