I recommend sticking with a syngeneic system. Only about 1/10 human lymphomas grow in SCID mice, so it gets very time consuming and expensive to do these experiments. The Levy lab at Stanford used A20 a lot, as do a lot of other labs around the country, so all of the conditions have been worked out and you will be able to jump straight into experiments.

There should be a number of cell lines from mouse (but maybe from different strains). You may check carefully on the ATCC.org website (keywords could be lymphoma, mouse, but you may make sure you don't choose lines from fusion experiments).

I do not remember all lines I have been working with, but I usually used them in cell culture experiments, not in mice (so I do not know how good they are transplantable).

So, you may check some cell lines: BW5147 , RAW.... (there are several probably very different ones with different numbers), WEHI ..... (different ones).

Some of the common lines were once generated by some mouse-retroviruses; some were homing to different organs due to cell adhesion receptors...

Thanks for the answer, it is a good advice, but I have already looked through ATCC and ECACC. There is a lot of cell lines with a potential, however, I would prefer to know which one for sure is transplantable as somebody used it and it works. PubMed is good but for that is a lot of work (and not all publications are full access).

I found this publication which uses MEC1 cells in a Rag2 deficient mice to model CLL.

So, I only shortly checked the one line I suggested in my first answer; I found at least one paper where it has been used to inject into syngeneic mice - and obviously re-used tumor-derived cells for injecting those again. I only have read the abstract, so, in principle, many of those cell lines could grow still in mice of the same strain.

I know that animal facilities try to make sure that a mouse or rat strain remains identical over decades, but I do not really know how much a cell line can differ from a strain after decades; I beleive that there could be some difficulties if a strain is extremely rare (like one of the rat strains I was once working with for questionable metastasis experiments...); but this problem should not be that big in common mice strains.

As far as I remember the BW cells are very round - I used them for some adhesion experiments...

Thank You both very much for the really good publications, especially the one with the metastatic and non-metastatic cell line (author is on a ResearchGate and hopefully will sent me a full-text). Pity that the AKR mice are a very challenging strain infected with retrovirus. I must ask our Animal Core Facility if it could be introduced into our SPF conditions. Or check if somebody rederivated the strain. Or if the retrovirus is transmissible. I will query Jackson Laboratory and check if it is feasible. So it looks like my idea of the project could be workable. Thank You very much for the help!

There are many different types of leukemias and lymphomas in human (and probably animal models). Perhaps, this is not that important for your scientific question (I don't know, I just assume), but in some cases it may not hurt to discuss some of your questions with a more specialized pathologists or hematologists from any university hospital to avoid any wrong planing of your animal experiments; for example, maybe you better look only for a model for leukemia rather than for lymphoma (or vice versa, depending on your scientific question, or, on how you want to measure progressing tumor or therapy approaches - this could be crucial at later stages of an time consuming experiment).

Thanks for the very good observation. As of now I just was thinking if the idea alone was feasible. And for that tumor cells need to be round or roundish. But of course, for the project proposal the application of this idea will be important. So, I will choose the kind of tumor which will allow me to get the best answers from both the in vitro and in vivo experiments. But I am also thinking about using both: leukemia and lymphoma model. Lately there is a big trend for the researchers to analyze in animal model different tumors to get to the common features of the tumor response or to make the generalizations in tumor response to therapy (just looking through the latest publications on PubMed). Two recenzents of my last publication were disappointed that only one tumor cell line was analyzed in vivo and in vitro. We will see. I still need to flesh out this idea a lot. And as I am working on a few ideas for the new projects at once, so thank You very much for the help with this one!

Hi, Patrycja, maybe this recent CELL-paper could offer you another model, using human cells and a cell line in SCID and NSG (nonobese scid gamma) mice. The advantage in that model is, one can test the therapy outcome, in this case a well established antibody in human NHL therapies against CD20, Rituximab, was combined with anti-CD47 antibodies - and leaded (in mice) to an interesting new cure of at least some cancers. Of course, it is more expensive and challenging to establish a SCID colony in the animal facility, but the options for developing new cures on human cancer cells (or testing on existing cell lines, I think they also used Raji cells) appears to be worth of trying. (I am not involved in any of such studies nor am I anyhow related to the patents or companies involved).

SCID mice are more expensive and challenging, however, You are right, this model gives more answers about clinical applications of the anti-cancer therapy than other animal models with syngenic tumor cell lines. And we have here, at the Medical University of Gdansk, a great animal facility fully prepared to work with such mice, so this idea has a great merit and is quite well workable. I should also add that the MAIN disadvantage about putting human tumors into SCID mice is that you eliminate the contribution of the anti-tumor immune response. Which, in my opinion, is the most important thing about in vivo models compared to in vitro cell culture.

Thank You for pointing out the problem with SCID mice and lymphoma. I have only worked with syngenic mouse models so far. I have looked at the publications by Ronald Levy (and You) about A20 lymphoma and thank You for directing me to them as they are very nicely written and interesting. And we already have BALB/c mice.

However, I am still thinking about checking one of the best working agents on human cells in vivo. I have plans about working in vitro on both human and murine cell lines analysing a group of drugs, than moving to syngenic mouse models (minimum two), getting the best in vivo working drug and then checking it in vivo on human cells in a few SCID mice. It would give a full picture.

As You work with lymphomas, maybe You have some idea if one of them have the better chance to grow in SCID mice? Would You give me some advice about that?

You have the best chance of a lymphoma growing in a mouse if it grows where it shouldn't in a human. For instance, a leukemic Follicular Lymphoma or DLBCL, or pretty much any primary lymphoma that you get from a pleural effusion.
