I am trying to work out the activity of trypsin in pancreatin using the TAME assay. I am following the Minekus et al method, detailed in INFOGEST (as this is a precursor to an INFOGEST I am setting up). However, this assay in particular seems to be causing a lot of problems. Namely, the pancreatin does not dissolve in the 1mM HCl. Furthermore, when running the 10min on the spectrophotometer, my results are quite bad, with really low readings (usually .030-.050) and show little difference from the blank. I was wondering if anyone else has followed this method and made any modifications, or has any suggestions on where I am going wrong?
