Neomycin sulfate (HY-B0470) is an aminoglycoside antibiotic that mainly acts on prokaryotic cells (such as bacteria).

Its antibacterial mechanism is to bind to the 30S subunit of the prokaryotic ribosome and inhibit protein synthesis.

However, it is difficult to penetrate the cell membrane in mammalian cells, and its efficiency on the ribosomes in eukaryotic cells is extremely low, so its cytotoxicity is weak, and it is impossible to effectively screen stable cell lines carrying neo resistance genes through selective pressure.

G418 (Geneticin, HY-17561) is a semi-synthetic derivative of neomycin, which enhances the permeability and toxicity to mammalian cells through structural modification.

And its mechanism of action is similar to that of neomycin, but it can effectively inhibit the function of eukaryotic cell ribosomes, so it has become the choice for screening neo resistance genes (encoding aminoglycoside phosphotransferase).

Different cell lines have different sensitivities to G418, and concentration pre-experiments must be done first.

First, use G418 to establish a concentration gradient (usually 200–1000 μg/mL) for preliminary experimental optimization to determine the lowest lethal concentration.
