First I want to state that I do not believe that any journal or journal group can guide any research. They are doing business, up to that it is OK. Practically a few people act as powerful reviewer decide what is research or what is not research. They have no authority to do so.

1. What is 'crude extract'?

Technically any solvent extracts of dry plant parts can not be considered as crude extract. But this term is used globally. I think it is wrong.

Only the direct extract of succulent plant parts (Juice) can be given that name.

2.Who said that multiple component of a plant juice can not be used as a medicine?

If it can be effective as a good medicine without toxicity, it MUST be considered as a medicine.

3. The standard Disc diffusion assay can be used for direct antimicrobial assay of plant extract. What is the problem?

Our Target should be research, not the journals.

4. Yes, many journal will not publish such paper. They fear losing of present medicine market from the Giant medicine manufacturers if any new type of medicine discovered.

I faced that problem personally.Then I convinced some like minded people and start publication of a new free, open access, peer reviewed journal which is totally open for any type of original research (www.animalmedicalresearch.org).

It is difficult to obtain such information, because these values ​​may be different for different groups of microorganisms (which are characterized by variable levels of sensitivity to substances).

The sensitivity to the extract was classified by the diameter of the inhibition halos as: not sensitive (–) for diameters less than 8 mm; sensitive (+) for diameters 9-14 mm; very sensitive (++) for diameters 15-19 mm and extremely sensitive (+++) for diameters larger than 20 mm

I hope experts and scholars continue developing new methods for evaluating the antimicrobial activity of plant extracts (juices!), which is totally different in mechanism than regular synthetic antibiotics.

I agree with Amel Amrani answer and others. However, this doesn't mean these classifications are always true, because due to the limitation of diffusion methods by itself. Larger inhibition zone doesn't always mean highly sensitive because in diffusion the lighter compound diffuse wider than heavy compounds and in another case the diffusion behaviors will also depend on the affinity of biologically active compounds to the medium. So that is the challenge working with crude extracts. It could have been easier for pure and chemically characterized active compounds.

Yes, plate-diffusion method is easier with pure compounds.

Also, if the nature of plant extract is hydrophobic,it is difficult to diffuse in agar medium which is a hydrophilic in nature.
