After fixing the cells with methanol, there are flocculent precipitates, which are not exfoliated cells.

The cells are not washed with PBS before they are fixed. Does the culture medium have any effect on the fixation?

Not the culture medium itself; it's the serum you add to it. If you wash your material with the culture medium alone, without added serum, you should not see the flocculation caused by the methanol.
