I am currently conducting experiment on determination of total flavonoid content by using Quercetin as standard. After reading few literature, I found out some researchers used 415 nm and some used 510 nm as maximum wavelength. My question is which wavelength should I follow and why? I noticed different wavelength used for different methods even though it was for same standard in this case Quercetin. For example, AlCl3-NaNo2-NaOH method used 510 nm while AlCl3-Methanol method used 415 nm. Someone please clarify this. Thank you.
