HI all

I work on yeast and I thing Ihave hyperglycosylation structure and I did the Con A and I saw my peaks in flow throw step not in elute, my Q that if the structure of sugar consist of hyperglycosylation or contain phosphate group can bind to lectin Con A? thans

Dear Hussein,

Yeast produce long, branched chains of mannoses (mannans) often linked to proteins. See here: http://www.ncbi.nlm.nih.gov/books/NBK1941/

These are perfect binding partners for ConA.

So if your protein is indeed glycosylated it should bind strongly to ConA.

If your protein is resistant to high pH try eluting it with unbuffered 20 mM diaminopropane. It should be stronger eluent than competing sugars.
