How can extract polyphenols from the leaves high in chlorophyll. I conducted an extraction from green leaves, I defatted the leaves with haxane and then extracted them with methanol but the green color indicates chlorophyll is still present.
I suggest that you extract the polyphenols in the solid phase extraction before measuring them. This method gives clear results and high phenolic values in the extracts

in my point of view it is practically impossible to totally remove the green colour from chlorophylls in extracts. It should be sufficient to defat the leaves (separation of waxes, lipids and also chlorophylls). By separation of the methanol extract and fractions thereof e.g. with Sephadex LH-20 column chromatography the green colour will no longer interfere with the isolation of the phenolic constituents.

One of the most often used methods for eliminating chlorophyll and other undesirable non-active pigments from methanol extracts is activated charcoal.
