I have a question for the culturing of MDA-MV-231. If I use complete L-15 (+10% FBS), can I cultivate the cells with 5% CO2? Because I saw that ATCC recommend the conditions without CO2 but it's more convenient for me to culture this cell with 5% CO2. Thanks so much for your suggestions.

I will not make any direct comment on ATCC recommendation of cell culturing like here culturing of MDA-MB-231 in l-15 medium and without CO2 condition.
But I am working on MDA-MB-231 from past 3 years. I am culturing MDA-MB-231 cells in DMEM+10%FBS in 5% CO2 incubator.
They are growing well and good without any issue.
I have not familiar with L-15 media, but I think if you used I-15 and 5% CO2, I am sure it wont affect cells.
Thanks so much Saurabh. What is the concentration of Glucose in your DMEM? Is the Glutamine also essential for this cell line?

I am using DMEM High glucose, obtained from Gibco (Cat#11965092) which contains of Glucose (4.7g/L) and L-Glutamine (0.5g/L) to which I add FBS (10%).
My personal suggestion, without any hesitation and complicated scenario you go with DMEM+10% FBS in CO2 incubation. MDA-MB-231culturing will work best without any issue.
