We are growing cells on 316L stainless steel. We are going to look for cell-substrate attachment using optical microscopy, but we also wanted to see whether or not the cells experience osseointegration. What are ways in which we can test for this?
Right now the only method that I can think of is to use a reverse torque test, but I wouldn't want to do that until the cells have spent at least a week or so on the steel surface.
Any tests for osseointegration that I can repeatedly do day by day?

animal experiments you can do removal torque. histomorphometry can actually quantify osteoblatic activity at bone implant interface. in humans you can check stability with RFA but osseointegration??
A recent definition (by Schroeder et al.) defines osseointegration as "functional ankylosis (bone adherence)", where new bone is laid down directly on the implant surface and the implant exhibits mechanical stability (also known as primary stability – i.e., resistance to destabilization by mechanical agitation or shear forces).
So i guess the definition itself should provide you the answer.
Visible attachment you are assessing via optical microscopy and functional stability can be assessed best by removable torque test. But doing that regularly/daily might have some effect on healing, if it moves even in microns. So i feel you can combine two techniques, select a parameter above which you'll start using torque test.
For e.g initial healing phase use RFA, after a certain acceptable readings are reached then you can use removal torque test in frequent intervals.
