Hi, I have a 25 ml sample of chitosan coated nanoselenium with average diametere of 112 nm, considering the fact that chitosan after being kept in water media swelles, how can I send a sample for TEM or SEM analysis? Can it be in water media (dispersed in water) or should I send dry samples?
For TEM, we generally use ethanol. Have you tried in ethanol?
in both cases the sample is tested in dry state.
For TEM, you can drop the dispersion of your material on the TEM grid and then dry the samples on the TEM grid.
For SEM you can test the powder or coating (by drop-casting) on aluminum foil.

I report our experience with silver nanowires and core shell nanowires (diameter around 100 nm). The wires are dropped on a mesh which is used for this investigations. The main problem is the transport in air. I remember that we dropped the Ag-nanowires on the mesh and then we waited 2 weeks over Christmas.
After 2 weeks the nanowires were completely dissolved due to the sulfur dioxid in air. So it is the best to transport the wires in a suitable solution (in our case it was pure water) and then prepare the mesh immediately before the measurement.
The other question is that the wires are fixed on a platform for other investigations. Than the platform must be suitable for TEM investigations.
