Does anyone have experience with a genome extraction kit/protocol that has worked well for T7 phage specifically? I’m looking to do whole genome extraction of T7 mutants for Next-Gen sequencing.
If you have phage, for example, in SM buffer (100 mM NaCl, 25 mM Tris-HCl pH 7.5, 8 mM MgSO4, 0.01% (w/v) gelatin), filtered through 0,22um filter, then it's a "mixture" of proteins and DNA, so you can make the simpliest phenol extraction. This method has been used a lot of times in my case, always fine.
Hi, I tried the method mentioned by Dr. Klimuk, after enzyme digestion, the DNA been extracted seems to be BL21 bacterial genomic DNA, which can not be cut in appropriate sizes. T7 phage needs to propagate in BL21 bacterial cells, so I was wondering how to separate the T7 phage genomic DNA and bacterial genomic DNA? Thanks a lot.
In my laboratory, we earlier used a method similar to phenol extraction proposed by Dr. Klimuk, previously modified by digesting E. coli DNA by DNAse and thermal denaturation of the phage proteins. This may solve the host DNA problems observed by Jiani Xing. At this moment, we use the commercial kit linked above.
