DMSO was reported to boost sensitivity in LC-MS analyses of peptides (PMID: 23975139) but I personally know couple of people that actually get worse results when they add 3-5% DMSO in mobile phases. What is your personal experience?

Hi, On my experience I can say that the DMSO ability to improve your mass results depends on the peptide charge And solubility in DMSO. For instance DMSO (0.1-0.5%) in water allowed to see a good ionization of some apolar 20mer peptideS. Finally I suggest you always to check both positive and negative ionizations on your spectrometer. Best 😊
Excellent question. We used 2% DMSO for untargeted metabolomics of lipidic molecules and generally got better s:n (must add: our motivation was target molecule stability and not s:n). However plenty of people have used DMSO in proteomics and at least one excellent study validate the results in HRMS quantitative proteomics (Strzelecka et al. 2015). We tried this shortly after the paper came out. It improved sensitivity for peptides a little but was overall a complete disaster.. we had to clean our Orbi Velos every other week (from approx. 4 months between cleaning without DMSO). We ordered and used the same DMSO described in the methods of the 2013 Hahne manuscript.
Thank you all for your contribution to the discussion. When we used DMSO in nanoLC-MS analyses we obtained less identifications than without it. However, when we used 3% DMSO in conventional-flow LC-MS the results improved significantly although we didn't see boosting sensitivity but rather the charge state coalescence effect (PMID: 29582996).
