I am performing a screen to evaluate the role of lipoproteins/cholesterol on gene expression, in which using lipid reduced/free FBS is essential. I have previously used Hyclone Lipid Reduced FBS SH30855.02 but unfortunately Thermo Fisher has discontinued this item. Any suggestions on substitutes from a reliable source? Thank you!
Gibco sells charcoal stripped FBS through ThermoFisher:

It is a bit pricey but if must have something like it, you have to either pay absurd prices or use activated charcoal column yourself to remove/reduce lipids and other non-polar material, followed by filter-sterilization.
You may also wish to consider defined medium, if your system permits its use.

I talked earlier with Thermo and according to them the Gibco Charcoal Stripped FBS reduces the levels of hormones and growth factors but does not reduce the lipid concentrations. So most likely, the cholesterol concentration will not be much different in the untreated and charcoal stripped serum and unfortunately it is not suitable for my screen..
Several companies sell serum free media for specific cell lines but I don´t know whether they add lipids to their supplement. Another possibility would be the preparation of lipid depleted serum by ultracentrifugation.

In defined media, in addition to amino acids, vitamins, salts etc. some components such as albumin and fatty acids are added to support cell growth. I am not sure about cholesterol but you can always call the tech service people from any number of suppliers and ask them. They may or may not be willing to give you the exact quantity of added cholesterol as they may consider it to be part of their proprietary information but they would at least tell if it is present.
If necessary, you can always quantify it yourself, it's not very complicated.
