I would like to know the recommended dose of streptozotocin(STZ) for induction of diabetic nephropathy and the duration required after (STZ) injection to develop diabetic nephropathy.
Also, are there any special precautions during the model induction

In my experience, 45-50 mg/kg STZ (dissolved in cold distilled water) have good results ( rat's weight, fasting before and after injection and volume of injection are important for success). According to evidence, it is well established, at least 3 weeks after diabetes confirmation the signs of diabetic nephropathy have observed in Wistar and Sprague rats. However, others factors such as hyperglycemia severeness have important impacts on this model. All in all, rodent model of diabetic nepropathy dont represent all aspects of the disease same as human. For example, the Kimmelstiel-Wilson lesion ( a pathological feature in end stage kidney diseases like nephropathy) dont develop in rodent model of diabetic nephropathy.

Thank you very much for your reply. but i would like to know more about the rats weight. Is there a recommended weight at the beginning of the experiment ?
The rat weights between 180-220 g are suitable.Induction of diabetes in rats with high weight (250) is difficult and with high rate of mortality. Moreover,in my experience, obese rats could reverse their hyperglycemia condition.
I really appreciate your help. I am also concerned about the survival of animals along 3 weeks for diabetic nephropathy to develop... are there any precautions to be considered during the experiment ?
