need to do an overnight E .coli culture from a colony or grow it to at least OD of 1 but can't do the next few steps for overnight induction - what happens if i store the culture in the fridge for a day or two? just doing an optimisation of my IPTG concs and testing what works best

It depends on the next steps, if you then dilute into fresh medium and let them grow a few hours before adding IPTG you should be fine. But I would not suggest to add IPTG directly to the stored cultures since you may not get reliable and reproducible results.

My suggestion is that the next day, Adjuct the OD 0.2-0.3 by using broth and keep it for 1-2 hours at 37'C till the OD will reaches upto 0.5-0.8, after that use culture for induction.
