for my project I am isolating EVs (such as exosomes and microvesicles) from mesenchymal stromal cell culture medium.
Because of the current situation in my country, I cannot immediately isolate EVs from the obtained culture-medium, so I will have to store the medium for a couple of months.
I will centrifuge at low-speed before storing, to deplete cells and large detritus.
My questions are:
-Do you have any recommendations which vessel I could use (ISEV recommends siliconized vessles) for around 150 ml of CM?
-Do you have any experience with keeping CM at -80°C prior to EV isolation?
-What supplements should I add for freezing at 80°C?
Hi,
I realise it's been quite some time since you asked this question about recommended EV storage conditions. We just published a comprehensive study looking into that, turns out the buffer used (and the purity of EVs) are more crucial contributors to sample loss than the tubes. We compared lots of different buffers at different temperatures, for up to 2 years of EV storage.
