Spheroplasts preparation for Yarrowia lipolytica?

I want to isolate lipid droplets from Y. lipolytica stationary phase culture cultivated in nitrogen limited minimal medium.

Does anyone have a cocktail mix recipe including zymolyase for Spheroplasts preparation??

Zymolyase treatment alone is not sufficient and I am losing LDs from intact cells!!
