Hi !

For the past 3 days I realized my iPSC did not attach that well after thawing and were detaching. Today I saw the formation of spheroids. Although there is still a lot of cells that attached and formed colonies (although they don't look great, especially on the edges). I also included what it looked like the day after thaw so a day before I saw full "spheroids".

The incubator, reagents, plates did not change. I have the same Matrigel aliquoted lot that I used for the past couple of months.

I just don't know what the issue may be... I had thawed a vial from the same lot once before and this did not happen.

I know that spheroids can form spontaneously but I am not sure what the problem is as I haven't changed anything. I am planning on changing incubators (as I am thinking the CO2 sensor might be faulty) and try out a different matrix (ECMatrix™-511 E8 Laminin Substrate | CC160-350UG).

I also realize that I might have mycoplasma (which I plan on testing), but would they form spheroids and still have relative good attachment like this?

If you have any experience regarding something like this happening, please let me know !
