NLCs were prepared by probe sonication technique. Prepation prepared after sonication was found to be clear with no sediments after centrifugation. Organic acid (medium chain length) was chosen as drug. Size of drud loaded NLC was around 30-50nm while that for blank was around 100-200nm.
Also product obtained after lyophization was in a paste form and not solid, is it acceptable?
It seems you broken the structure of NLC by removing the liquid oil fraction. Now you probably have a mixture of drug loaded polymer nanoparticles and an emuslion of oil. That's why after the lyophilisation material is in a paste form. Maybe you just oversonicated it.

Following sonication conditions were followed:
Sample overall run time for 15min with 59sec on and 5sec off period at 40% amplitude maintaining the temperature around 60-80 oC.
Kindly convey if any alteration to the above procedure can be applied.
Dear Deepinder, try to maintain temperature below 40 C. It should be done by varying on/ff cycles timjng or by reducing the amplitude.

Dear Deepinder, you can change the crosslinking agent and try it again with controlled temperature and sonication speed.
Check the compatibility of organic acid you choosen with polymer.
May be you should use some other technique to form NLC as sonication may be a harsh technique for NLC formation. What method you are using to do the particle size analysis. What is your PDI?
Dear Deepinder, do you have checked the physical state of the encapsulated phase by DSC. If the core media is liquid the vesicle sizes are influenced.

It seems there is some physico-chemical interaction between your drug and the lipid used for preparation of nano-structured lipid carriers.
I think you should study this interaction first through performing some experiments for example DSC, XRD or FTIR which suits your drug and polymer structures.
The bigger size of your blank NLCs compared to your loaded NLCs also the unstable freeze-dried product might confirm my assumption.
I totally agree with previous replies offered by other colleges regarding changing the preparation conditions.
Particle size is analysed using Malvern Zeta sizer with PDI values ranging between 0.2 to 0.4.
