There are 7 lysines in my protein molecule. I would like to specifically biotinylate a lysine residue in my protein. How to block other lysines from being biotinylated? Is there a simple way to do that?
I am afraid that there is no way to do that, let alone a simple way, if the lysines are randomly distributed across the sequence of your protein. One of the lysines will probably be the preferential labeling site, but it will be difficult to label only that to a high degree without also modifying other lysines.
You could consider using a type of labeling that is specific for the N- or C-terminus of your protein, if you'd like a relatively homogenous singly labeled pool.
