I'm on my way to do my research in analyzing Ascaris suum body fluid. I have several questions, since I couldn't find the answer everywhere.
1) To wash the Ascaris suum after isolating it from pig intestines, should I use NaCl 0,9% or PBS 4x? The PBS 4x I got from previous literature, and I was asked to consider using NaCl 0,9% since it's cheaper and it doesn't have any direct contact to body fluid.
2) I have to dilute the body fluid of the worm. Should I use PBS, NaCl 0,9%, or aquadest to dilute the body fluid? First, if I use aquadest, it should destroy the cells inside the body fluid and may change the molecules contained, should it? Second, if I use NaCl 0,9%, it should give change to the ultrastructure of cells. But, since what I'd like to analyze is the protein, will it give some effects? Third, If I should use PBS, what kind of PBS should I use (1x, 2x, 4x)? They differ in molarities, so I think there are some differences in effect.
