I am about to test the anticancer activity of my plant extract on a cell line. Where should I dilute my plant extract? 0.1% DMSO then add it to the cell media? Or directly with the cell media?
In my opinion, both can be used but I would prefer cell media instead of DMSO. It is a bit dependent on the solubility of the plant extract. Which solvent or mixture was used during extraction? This affects the Phyto molecule diversity at your final crude extract and different polarities may result in different solubility behavior. DMSO may improve some hydrophobic phenolics and mentioned concentration is not toxic for the cell. If the organic-free fully aqueous recipe was used as an extraction reagent DMSO is not needed, the most convenient will be the medium.
