In our lab we investigate the biofilm formation of uropathogenic E. coli and I would like to perform a random mutagenesis experiment to be able to identify genes mediating this process.
However, I lack any suitable screening assay to select those mutants with defective biofilm formation. Because I am actually pursuing a negative phenotype (the lack of biofilm formation) I find it hard to conceive a suitable screening for this case.
Could anybody suggest an elegant screening method to identify biofilm-defective mutants?
