I want to prepare a termite sample for SEM observation and i know it is dehydrated with a graded series of ethanol. but my sample has already been fixed in 75% ethanol for more than one month. So how to treat these samples to prepare for Scanning electron microscope? kindly suggest.
I suppose you need to observe only chitin shell of a termite. You can place your insect in 95% ethanol overnight, then - 100% ethanol (dehydrated) overnight, then submerge it in HMDS for an hour and let it air dry in a fume hood. If you do not have dehydrated ethanol and HMDS, you may want to try conventional (94-95%) ethanol (overhight) and air dry for two days. It may work.
