I extracted some plant proteins using a phenol-based method and then precipitated them with ammonium acetate in methanol. I need to quantify total protein using the Lowry method (absorbance at 750 nm).
Laemmli 2x sample buffer interferes with the Lowry assay, leading to inaccurate and inconsistent readings. Unfortunately, I don’t have access to the Bio-Rad DC Protein Assay kit, which includes Reagent S to handle detergents.
Does anyone have experience with a homemade or alternative buffer that is compatible with the Lowry method and still allows full resolubilization of proteins extracted via phenol?
Any buffer recipes or tips would be greatly appreciated. Thanks in advance!
