I am going to construct my own cassette which will be Dox inducible, i came across several names of tetracycline activator (rtTA). upon sequence alignment i found some differences. For example, rtTA3 is of 708bp and rtTA advanced is 747bp. rtTA3 has shorter VP16 transcriptional activation domain, and i have not checked whats in M2rtTA has. Similarly TREGV promoter and Tight TRE promoter have also some sequence dissimilarity, for instance lack of CMV minimal promoter. I wants to know which Dox inducible system is updated and have shown better induction? i am going to take this cassette in safe harbor region, maybe AAVS1 locus or ROSA26.

(please refer a plasmid with addgene number, so i can directly take the sequence from it)
