I need to extract RNA at OD(650nm) 0.1-0.15. With this OD, harvested pellet is too too small.
I am following Nucleozol protocol.
Earlier, for 0.5-0.7 ODs, i was using 20ml and was getting decent concentration of RNA.
But now there are 2 points to be considered: Low OD and Min Media is being used.
So my questions are:
1. how much culture volume?
2. tips to increase RNA concentration (as for the last 4-5 weeks, i am constantly getting low concentration).
3. tips to maintain its integrity.
