I would like to know if the retroviral transduction on Mesenchymal Stem Cells is better option that lentiviral transduction. We need to inhibed the expression of some proteins of TLR3 signaling pathway in this cells and we know that MSCs are dificult to be transfected.

I've used lentivirus (pVSVG with pCMR delta-8.2 packaging) to transduce human MSCs for iPSC generation. The GFP controls show extremely high transduction efficiencies (>95%). I also used this system to transduce a wide variety of cell types from different species. That was a long time ago, and I'm sure things are even better now.
Generally, lenti better at transducing non-dividing, or slower cells.

in our experience, Lenti-gets stably incorportated in the cells, better chances to retain your transgene, but Retro- can lose with time.
retro - can have issues getting incorporated into the genome. Hence go for lentivirus..but some labs prefer retro for mouse.
