I'm currently analysing some data on the effect of an agonist on calcium flux when coverslips of cells were separately transfected with different plasmids.

I'm using a 2 way ANOVA with the response to the agonist alone, the response to the agonist in the presence of an antagonist and the response again after antagonist washout being the column factors and the two plasmids being the rows factor. Whilst having the column factors as repeated measures makes sense as the measurements are from the same cells, because the cells are from the same original source before being placed to grow on the coverslips - and therefore not entirely independent -, should I also set the row factor as repeated measures?
