I have a sample of glycans which were removed via enzyme (this enzyme is glycerol-free), however my NMR spectrum showed a ton of glycerol. I want to recover the glycans, but remove the glycerol. How would I go about doing this with the greatest recovery amount? Also, if there is a method that quick (less that 48 hours)?
Edit: not sure where the glycerol came from, still trying to back-track where it might have come from.
How big are the glycans you're interested in? If they're large enough, you could dialyse through a low MWCO dialysis membrane to get rid of the glycerol.
As for where it came from, is it possible your glycan-binding enzyme has some affinity for glycerol too? If there is any glycerol in the samples the glycans came from originally, maybe the enzyme carries it through.
Philip Kitchen: Currently trying to back track where the glycerol came from via LC/MS. And we are currently looking into centrifugal MWCO filters (possible min size is ~1kD, give or take)
Good luck! One thing to bear in mind is that some of those filters are made of cellulose - potentially a problematic contaminant if you are studying glycans.
