It is logic for appearance lower IC 50 in anti-inflammatory assay (COX I, II & LOx) than standard/ control even though it has lowest in Total phenolic and flavonoid content And high IC 50 in DPPH and ABTS and vice versa, sample, has high total phenolic& flavonoid content with low IC 50 in DPPH and ABTS, gives high IC 50 in anti-inflammatory assay??
Dear Yasmin,
There is not link between DPPH, ABTS and anti-inflammatory assay. So you have to evaluate first all the activities before giving a conclusion.

I correlated that since the total amount of phenols and flavonoid is high, this means that they have high antioxidant activity, and as a result, IC 50 in the anti-inflammatory is low
but if no correlation between DPPH, ABTS and anti-inflammatory, but there is a relationship between total phenolic, flavonoids and anti-inflammatory (IC 50)

In general high content ph.and flav. in a bioactive compounds lead to high antioxidative and anti-inflamatory activity and have direct relationship to decrease IC 50 .

What was method did you use to measure anti-inflammatory assay (COX I, II & LOx) in vitro (using cell line or not) or in vivo (using an animal model)?
What kind of sample was that, crude extract or fraction?

Since you were using the in vitro assay to measure IC50 anti-inflammatory, the things you mentioned above could happen. This is because the antiradical activity (DPPH and ABTS assay) of your test material might not be directly related to its anti-inflammatory activity. In in vivo study, the flavonoids and phenols play indirectly as anti-inflammatory activity, they are enhancing nuclear transcription factor oxidative stress-related defense such as Nrf2 and also inhibiting the activity of pro-inflammatory transcription factors such as NF-kB. Other compounds such as tannins contained in the test material might have a direct impact of inhibiting the activity of COX I, II & Lox so that pro-inflammatory cytokines are not formed.
