Hello Everyone. I was trying to label antibody with Cat# MO-L007 - Cystein reactive monolith series antibody labelling kit. I followed every steps that the protocol states. But in the final step after labeling with the blue dye, while i let it pass through column B-AB, I got the antibody as transparent and not at all even light bluish. What I saw is that blue dye was stuck in the gel of the column. If the antibody is not even light blue/blue, then how can I measure the absorbance using Protein Lowry 650nm in Nanodrop? It will be great help if you please suggest me the needful.

I am not familiar with that labeling, but generally anything that reacts with cysteine may denature the antibody. I would use something that labels free lysine residues. Even then you will have to determine if the labeling affects the function of the antibody.
