I have read some articles suggesting that acetaminophen could have some psychotropic effects such as antidepressive effect with some patients. Do you agree with that? are there any patient related factors such as genetic factors that enhance this effect?
One of mechanisms included in acetaminophen spectrum of action is blocking of the reverse transport of endocannabinoids. What do you think abuot such explaination ?
Thanks Przemyslaw Zakowicz , that mechanism could be contributing to the antidepressive effects of acetaminophen, what about the role of genetics? based on that mechanism of action, does acetaminophen have any kind of substance abuse? if you have more explanation, that would be appreciated.
I am afraid that also in this case, like in whole psychiatry, genetic factors determine only 'vulnerability' to an effect (e.g. receptor gene polymorpsismh, CYPs et c.). Going further towards cannabinoid hypothesis, antidepressive effect could be an effect of depolarisation- induced suppression of inhibition, resulting in dysinhibition of neural activity, moreover, animal models of depression use fear - conditioned learning to induce mood decrease- if cannabinoids modulate synaptic plasticity (disturbing of LTP) maybe they can also block rebuilt of neural network, triggered by fear and stress. Thats my opinion, or rather hypothesis.
