Does anybody have a protocol for keratinase assay, using keratin or azokeratin as a substrate?
There are many methods available for keratinase assay starting from simple caseinolytic method to little bit complex keratin hdyrolysis methods; it depends on your lab feasibility and possibility. If u supplement only keratin source as C/N and energy source in the medium along with test strain, post degradation, u can directly measure the peptides/amino acid equivalents available in the culture supernatant by a simple protease assay too. Your purpose of assaying can decide your method of selection. For ex, just to check feather degradation activity of XYZ strain, u can follow the caseinolytic assay; but for further confirmation and all that u need to go through keratinase assay where we will determine our enzyme capacity whether it can hydrolyze keratin or not..
Keratinase assay can be carried out by two means; it may through UV methodology or visible methodology......
