I feel like I have tried everything and wanted to reach out to you all to see if anyone has any suggestions for staining the dentate gyrus of my 40 um mouse tissue with DCX at 1:500. I have tried using H202, a pretreatment with .1 M Tris and .3% Triton, as well as .01 M Citric acid (pH 6) antigen retrieval for 10 minutes at 90 degrees. I have used different combinations of these steps as well as them individually and have an overwhelming amount of background and feel as if there is nonspecific staining occurring.
