hi i really need advise on which proliferation assay to choose from.
my experiment involve growing cancer cells on transwell insert with pores of 0.4um that does not allow invasion. After confluency is reached at about 50%(1.5 days), i intend to apply a weight over the cell, putting compression onto cell. The compression duration is 16hrs approximate. Which proliferation assay is best for this condition?
i have thought about BrdU assay but one protocol i read described exposing the cell with BrdU for one hour only. Since my treatment time is about 16hrs, this does not seem suitable?
Also it is important that the assay is able to quaantify proliferation only during the 16hr of compression.
