Hello! I am working on the extraction of alkaloids from a plant. I prepared a degreased hydroalcoholic extract, to which I performed an acid extraction and then basified. This last basic fraction I have tried to extract it with a series of little polar solvents but I am unable to extract the alkaloids from the aqueous phase (the reaction with Dragendorff reagent continues to be positive), it seems that the alkaloids present are very polar in nature.
Any recommendations on how to extract them from the aqueous phase?

No need for their extraction from the aqueous phase. Its better to set them dry and start their purification using column chromatography, reversed phase silica and water : methanol gradient mobile phase
Since its still in the aqueous phase, try a more polar solvent like n-butanol using a separatory funnel. Shake very well and allow to stand for some minutes. Then collect the organic phase, concentrate this phase and use column chromatography.
